The Role

We are seeking a motivated postdoctoral Research fellow to join our pluripotent stem cell group at the Newcastle University Bioscience Institute to develop hypoimmunogenic photoreceptors for treatment of advanced retinal degeneration.

The eye is suitable for transplantation so replacing dysfunctional photoreceptors should be feasible as long as sources of such cells are readily available. Pluripotent stem cells (PSCs) can generate cone and rod photoreceptors, which are able to integrate into the host retina and rescue vision in pre-clinical models of advanced retinal degeneration, but efficient long-term integration of allogeneic PSC-derived photoreceptors is impeded by the host immune response. To overcome this barrier and make this treatment accessible to a large number of patients with photoreceptor degeneration, we propose to generate off-the-shelf, immune-transparent (hypoimmunogenic) PSC-derived photoreceptors.

This ERC/EPSRC funded award follows from our recent work on characterisation and transplantation of CRX-photoreceptors in rd1 mice (PMID: 30681766) and measuring their light responses (PMID: 33657251).

A scientific background on pluripotent stem cell culture and differentiation to retinal organoids and retinal biology will be essential and applicants with record of accomplishment and expertise in gene editing in pluripotent stem cells and retinal organoids are preferred. Importantly experience of immunofluorescence analyses, flow cytometry and immunology-based assays are highly desirable for this post.

The post-holder is expected to take up this post from 1st of March 2024 fulltime for 3 years on a fixed term contract.

As part of our commitment to career development for research staff, the University has developed 3 levels of research role profiles. Key Accountabilities
- Use CRISPR/Cas9 gene editing to generate hypoimmunogenic pluripotent stem cells
- Perform the differentiation of human pluripotent stem cells to retinal organoids and their full characterisation
- Investigate the interactions between the pluripotent stem cell derived photoreceptors and those of the immune system in vitro
- Characterise the retinal degeneration phenotype in a new model of advanced retinal disease using immunofluorescence analyses and MEA recordings
- Regular liaison with the Investigators and researchers working on retinal and stem cell differentiation team and collaborators to ensure high quality of research and timely delivery of the project goals
- Perform data extraction and analysis and ensure full compliance with existing ethical regulations as appropriate
- Lead on the production of high-level scientific information including analysing, writing and presenting reports and manuscripts relevant to the transplantation work
- Continuously review the literature and technology relevant to the project and ensure that the outputs of the project are positioned appropriately in relation to other on-going work
- Supervise BSc, MRes and BSc project students

The Person (Essential)

Knowledge, Skills and Experience
- Knowledge of retinal development
- Experience in generation of retinal organoids from pluripotent stem cells
- Experience in retinal tissue processing and immunohistochemistry or RNA-Scope
- Experience in situ gene editing in pluripotent stem cells
- Experience in flow cytometry and Elisa-based assays

Desirable
- MEA recordings and data analyses
- Work with mouse models of retinal degeneration
- Broad knowledge of immunology
- Evidence of a record of research including publication and presentation of data at conferences
