The Role

We are seeking a motivated Senior Research fellow to join our group of investigators (Lako, Armstrong, Korolchuk and Steel) at Bioscience Institute to investigate choroid endothelial cell loss in Age Related Macular Degeneration (AMD) using a patient specific iPSC model. The Senior Research fellow will generate the control and patient specific choroid endothelial cells and assess the role of complement activation on their survival and transcriptome. Further the senior research fellow will perform pharmacological screens to rescue choroid endothelial cell loss in AMD patients. This project is funded by Macular Society UK and follows from our recent work on generation and characterisation of AMD specific RPE cells (PMID: 36544284, PMID: 32815311, PMID: 28913923) and single cell RNA-Seq of AMD choroid and RPE cells (PMID: 36645183).

A scientific background on pluripotent stem cell culture and differentiation to retinal organoids, RPE cells or choroid endothelial cells, immunofluorescence analyses of mouse retinas and retinal biology will be essential and applicants with record of accomplishment and expertise in complement biology and pharmacological drug screening would be advantageous.

The post-holder will work under the direction of investigators from September 2023 fulltime for 2.5 years on a fixed term contract.

Key Accountabilities
Perform the differentiation of human pluripotent stem cells to choroid endothelial cells
Fully characterise choroid endothelial cells using a combination of immunofluorescence, Elisa and western blot-based assays
Assess the role of complement activation in choroid endothelial cell loss in AMD patients
Perform pharmacological screens to rescue choroid endothelial cell loss in AMD patients
Lead the team of researchers involved in this project with respect to experimental planning and co-ordination of various assays performed in collaborating laboratories
Regular liaison with the Investigators and researchers working on retinal and stem cell differentiation team and collaborators to ensure high quality of research and timely delivery of the project goals
Perform data extraction and analysis and ensure full compliance with existing ethical regulations as appropriate
Lead on the production of high-level scientific information including analysing, writing and presenting reports and manuscripts relevant to the transplantation work
Continuously review the literature and technology relevant to the project and ensure that the outputs of the project are positioned appropriately in relation to other on-going work.

The Person (Essential)

Knowledge, Skills and Experience
Knowledge of retinal development
Experience in generation of retinal organoids, RPE or choroid endothelial cells from pluripotent stem cells
Experience in tissue processing and immunohistochemistry or RNA-Scope
Experience of molecular assays (RNA-Seq, Elisa, western blots)
Experience of setting up pharmacological screens and respective data analyses

Experience of lab management
Evidence of an excellent record of research including publication and presentation of data at conferences
