The Role

We are looking for an experienced researcher in cancer biology for this exciting opportunity. You will be part of a team based in the Newcastle Cancer Centre, in the Cancer Research UK Newcastle Drug Discovery Unit and will focus on identifying and validating new drug targets for oncology treatment, working within a large multi-disciplinary team that includes biologists, structural biologists, and chemists.

The project involves RNAseq analysis and data interpretation, immunohistochemical and microscopy methods, manipulation of selected drug targets using genetic or pharmacological strategies in tumour cell line models, and an assessment of the consequences of inhibition using signalling pathway readouts and phenotypic assays.

You will possess a PhD in cell biology and have experience in handling and analysing clinical and preclinical samples, using techniques that enable the ability to design, implement and deliver novel strategies to reveal drug targets within tumour cells and the tumour microenvironment.

This is a full time fixed term post tenable for 12 months.

Key Accountabilities
- Although working under the general guidance of an academic or Principal Investigator, the postholder will contribute ideas, including enhancements to the technical or methodological aspects of their studies, thus providing substantial 'added value'
- Handle clinical samples with full adherence to local and national ethics legislation
- Processing of clinical and preclinical samples for RNA, DNA or protein analysis
- Perform transient and stable transfection of cell lines. Familiarity with the design and use of genetic methods for target manipulation (siRNA/shRNA and CRISPR/Cas9) is required
- Use of cloning strategies to design and express mutant target proteins in cells
- Measure the expression and activity of proteins of interest in preclinical models and clinical samples, using western blotting, flow cytometry, ELISA, immunohistochemistry and other techniques
- Measure the expression of mRNA in preclinical models and clinical samples, with and without treatments. Previous experience of generating and analysing RNA datasets (including RNAseq) is required
- Interrogate proprietary and publicly available datasets of mRNA expression, DNA copy number, mutations and re-arrangements, from both clinical samples and tumour cell lines, to examine target expression in disease and assist the selection of appropriate model systems
- Use tissue culture to quantify endpoints (cell proliferation/survival, apoptosis, functional readouts) and have the ability to develop and use additional phenotypic and mechanistic assays to investigate the effect of target manipulation on tumour cell biology
- Carry out meticulous recording, storage and archiving of data, and perform statistical and data analyses to produce robust and reproducible data
- Present results at internal and external meetings and conferences or through publications in reputable outlets appropriate to the discipline
- Contribute to the writing up of their research for publication and dissemination, either through seminar and conference presentations or through publications
- Assist in the supervision of undergraduate and postgraduate students in the laboratory
- Contribute to laboratory meetings e.g. problem solving, internal seminars, journal club
- Perform risk assessments and maintain safety laboratory records (e.g. COSHH) in consultation with academic staff
- Organise and assist in general laboratory duties as required (e.g. purchasing, equipment maintenance)

The Person

Knowledge, Skills and Experience
- Use of gene-silencing and gene-editing methods for target validation purposes
- Expertise in mammalian cell culture, Western blotting, PCR, transfection methodologies, construct design, subcloning, site-directed mutagenesis, the analysis of mRNA and genomic datasets, and immunohistochemistry (with image analysis)
- Experience in the handling and processing of human clinical material and preclinical samples and the subsequent analysis of RNA, DNA, and protein endpoints
- Expertise in RNAseq analysis including single cell methods
- Experience in bioinformatic approaches including use of R
- Cancer post-doctoral research experience in an academic or industry setting
- Experience in designing and delivering research proj
