The Role

We are seeking a motivated Research fellow to join our pluripotent stem cell group at Bioscience Institute to assess new therapies for patient with Retinitis Pigmentosa (RP) caused by mutation in the pre-mRNA splicing factor 31 (PRPF31). RP is a common form of hereditary, progressive sight loss: it has a prevalence of 1 in 2500 births and 1 million people are affected worldwide. A major form of RP is caused by defects in components of the “spliceosome”, an important and complex structure within cells. The spliceosome edits unwanted or nonsensical passages out of a set of genetic instructions so that only intelligible code remains for the cell to use, but it is unknown how this links to eye disease. One of the most common causes of RP is a fault in a group of genes that regulate this process. We have generated retinal cells from patients with mutations in a key gene involved in the splicing process (PRPF31) and have demonstrated that retinal pigmented epithelial (RPE) cells and photoreceptors are affected at the structural and functional level. Here we aim to augment the expression of PRF31 using a gene therapy approach and asses its efficacy in restoring RPE and photoreceptor function.

This study provides a unique opportunity for rapid proof-of-concept (POC) studies, leading to the potential for rapid translation into a Phase I/II clinical trials for PRPF31 RP patients as an immediate outcome of our proposed research. The project involves close collaborations between our group at Newcastle University, with Prof. Robin Ali’s group at King’s College London, Dr. Sina Mozaffari-Jovin’s group at Max Planck Institute in Gottingen (Germany) and Dr. Sushma Nagaraja-Grellscheid’s group at the University of Bergen (Norway).

This project is funded by the Retina UK and follows from our recent work in this exciting field (PMID: 30315276; PMID: 35297555).

A scientific background on cell culture and differentiation will be essential and applicants with record of accomplishment and expertise in stem and/or retinal biology are preferred.

The post-holder will work under the direction of all three investigators from 1st September 2023 full time until 31.05.25 on a fixed term contract.

Key Accountabilities
Differentiate control and PRPF31 pluripotent stem cells to retinal organoids and RPE cells
Assess the impact of PRFP31 AAV augmentation on retinal organoids and RPE cells using molecular, electrophysiological and biochemical assays
Regular liaison with the Investigators and researchers working on retinal and stem cell differentiation team and collaborators to ensure high quality of research and timely delivery of the project goals
Perform data extraction and analysis and ensure full compliance with existing ethical regulations as appropriate
Lead on the production of high-level scientific information including analysing, writing and presenting reports and manuscripts relevant to the transplantation work
Continuously review the literature and technology relevant to the project and ensure that the outputs of the project are positioned appropriately in relation to other on-going work.
