**Background to the Project**

Chordomas are a rare, slow growing malignant bone tumour of the spine which are difficult to detect at early stages1. Patients are often diagnosed by tissue biopsy once a significant tumour mass has established with surgical intervention often the main therapeutic option. A new methodology for early diagnoses of these patients is urgently required to allow control and delay of tumour growth before surgical intervention.

Tumour specific extracellular vesicles (TsEVs) are a promising biomarker for cancer detection. EVs are formed from a phospholipid bilayer containing cargos of proteins, nucleic acids, and metabolites, which are important in normal cellular cross-talk. TsEVs have been identified in a range of cancers with altered cargos, leading to drug resistance, growth and metastasis of surrounding cells2. Detecting TsEVs in the peripheral blood of patients represents a non-invasive early diagnostic tool for tumour detection3.

Working with clinical experts at the Royal Orthopaedic Hospital, this multidisciplinary project will use research expertise in EVs, stromal cell biology and bioengineering at Aston University to explore the potential of TsEVs as a biomarker for the diagnosis of chordomas. Exploring and comparing vesicle composition from tumour biopsies to isolated chordoma and healthy cell lines will be investigated for a potential chordoma TsEV signature. TsEVs will also be assessed for pro-tumorigenic effects on healthy mesenchymal stem cells. Importantly, patient peripheral blood will be investigated for the presence of chordoma specific TsEVs, revealing the potential as a novel biomarker for diagnosis.
