AAV is a life-threatening multisystem disease characterized by necrotizing inflammation of small blood vessels and deleterious organ damage. Early disease control by targeted treatment has improved outcomes considerably, but the most important clinical challenge now is the recognition and control of flares. This project aims to delineate the immunological basis of disease flares and disease persistence in AAV patients. We have observed that individual AAV patients (in contrast to healthy individuals) can harbour large populations of B cells expressing IgM-ANCA. Based on this preparatory work, we hypothesize that defined auto-reactive B cell responses reflect a so far undetermined layer of immunological disease activity in AAV, with IgM B cell responses driving flares. To test this novel hypothesis, the project unites three expert centres that combine unique expertise and technology in autoreactive B cell biology, well-defined patient cohorts with longitudinal follow-up and biological samples, and patient representatives experienced in supporting translational research. The expected end-product is an immunological definition of (imminent) disease flares in AAV and a novel measure of disease activity.

**Duties and responsibilities**:
You should take initiative in the planning and execution of your research, including to identify and develop suitable techniques and analysis to deliver outputs that fulfil the requirements of the grant objective. You should always ensure the validity and reliability of your data, including good record-keeping. You should be willing and able to work both independently and as part of a team with a common goal. You should be prepared and capable of communicating your work, including in written reports and papers. Collegiality and maintaining a harmonious work environment are paramount.

**Essential requirements**:
You should hold a PhD in immunology, or a related field. Previous experience in flow cytometry is essential. Good interpersonal skills are essential.

We value careful documentation and meticulous experimental work, and we expect that you will have excellent organisational and communication skills. You will be joining a collaborative research environment with several other basic, translational, and clinical research groups focusing on inflammation, immunology, and haematology - and so a collaborative mindset and interest in team science is key.

**This role is full time, fixed term for 18 months.**
