Laboratory Student Placement in Metabolic Health

This is a 40 week fixed term student placement opportunity.

About the Role

This placement will investigate the role of zinc finger homebox-3 in mouse adipose tissue and cultured mouse and / or human (pre-) adipocyte cell lines.

Previous work in a mouse model with a single copy of a coding mutation in ZFHX3 demonstrated lower body size and fat mass with this mutation (PMID: ), this mutation is similar to a GWAS identified human missense mutation in the same exon of a highly conserved region of this gene (PMID: ). The Dumbell lab has been following up on the hypothalamic effects of ZFHX3 to regulate growth axis components in the arcuate nucleus to drive appetite and metabolic phenotype in mice, and the molecular mechanisms driving these changes in neurons.

However evidence exists for a direct effect of ZFHX3 to regulate adiposity in adipose tissue in flies (PMID: ), and studies in other cell types implicate ZFHX3 in JAK/STAT signalling regulation (PMID: ). This project utilises banked tissues from ZFHX3 mutant mice for investigation of mRNA and protein expression changes and phosphorylation status, as well as cell culture knock-down and over-expression experiments. The placement student will gain experience carrying out routine lab work in the active Dumbell research group ), working on cell culture and tissue extraction experiments investigating the role of transcriptional regulator ZFHX3 in adipose tissue / cultured (pre)-adipocytes.

You will gain valuable experience in an active and vibrant research lab, carrying out RNA extractions, cDNA synthesis and qPCR experiments (including analysis), protein extraction and western blotting, and routine cell culture of mouse and human (pre-) adipocytes.

The ideal applicant will already have some laboratory-based experience in one or more of the techniques mentioned, will have excellent communication and record-keeping skills, strong attention to detail, and be confident working independently as part of a busy research team.
