TheTriantafyllou lab () investigates the microbial, molecular and cellular crosstalk that underlies immune dysregulation during acute and chronic liver injury. To determine this, we study both human tissue samples and in vivo disease models and employ flow cytometry, cell culture, single-cell and spatial multi-omics. Throughout our work, we seek to advance our immunological understanding of liver disease pathogenesis and identify targets for developing immunomodulatory therapeutic approaches. One part of our research focuses on the dysregulated, and often ineffective, immune response to infection during acute (., paracetamol overdose) and chronic (decompensated cirrhosis) liver disease, that contribute to increased morbidity and mortality. Another significant area of interest within the lab is determining the mechanisms through which different checkpoint pathways [., PD-(L)1, TIGIT] control immune responses, at steady state and in liver inflammation, with the aim of exploiting this to produce new immune-directed therapies.


The post will focus on studying checkpoint pathway-mediated immune dysregulation in chronic liver disease and alternative therapeutic immunomodulatory approaches. The scientist will be based in the Section of Hepatology and Gastroenterology and work alongside the PI, lab members and collaborators [Prof Prakash Ramachandran (Edinburgh), Dr Mark McPhail (King’s College London)].

The post holder will generate transcriptomic data from in vivo models and human samples to perform scRNA-seq relevant analyses which will be combined with tissue multiplex immunofluorescence and flow cytometry. We anticipate that combination of these approaches will provide novel insights into the immunobiology of checkpoint pathways and will improve our understanding of their control of liver injury and fibrosis, and regulation of immune cell responses in the liver microenvironment. The ideal candidate will have good knowledge of immunology and computational background with experience in genomic data analysis and is keen to develop their skills and knowledge in the rapidly moving field of liver immunology and multi-omics. The successful applicant is expected to hold excellent communication skills and a genuine enthusiasm for exploring the interface between computational and experimental immunology.


Conduct research studying immune dysregulation in liver disease using human tissue samples (., blood) and/or in vivo models of liver injury. Plan and carry out wet-lab work (., immune cell isolation, flow cytometry, tissue multiplex stain imaging, microscopy, multi-omics).Conduct computational data analysis - apply/develop established and innovative computational approaches for the analysis of different multi-omics datasets (., single-cell RNA-seq or ATAC-seq, cell-type identities, ligand-receptor interaction modelling, spatial transcriptomics).Work collaboratively with both experimental and computational scientists to address complex and diverse biological problems in the liver immunology field.Assist with the data collection/analysis and ensure the validity and reliability of data.Development and maintenance of study design and standard operating procedures (SOPs)Undertake any necessary training and/or development courses.Provide guidance and supervision for student projects and/or instruction of students in the use of equipment, demonstration of techniques, and bioinformatic analysis approaches.
