As part of our multidisciplinary team, you will develop mammalian plasmid constructs for optimal expression of antibody therapeutics in mammalian bioprocesses.

About the Role:

The successful candidate will be responsible for designing and screening mammalian gene expression plasmids for high and stable expression of therapeutic antibodies. They will also design and optimize CRISPR knock-out and ORF libraries for high-throughput functional genomics pipelines.

Key Responsibilities:

  • Design and screen mammalian gene expression plasmids for high and stable expression of therapeutic antibodies
  • Design and optimize CRISPR knock-out and ORF libraries for high-throughput functional genomics pipelines
  • Develop complex R&D experiments to ensure timely deliverables
  • Document research findings clearly in a lab book
  • Communicate regularly in group meetings and prepare presentations for project teams and stakeholders
  • Contribute to laboratory maintenance, including equipment upkeep, cleaning rotas, and consumables stock management
  • Maintain laboratory health, safety, and quality procedures
  • Select, follow, and adapt experimental protocols and generate laboratory SOPs

Requirements:

  • BSc/MSc in Biochemistry, Molecular Biology, or related field
  • BSc/MSc with 3+ years of industry experience or PhD with 1-2 years' experience
  • Extensive experience in designing, preparing, and screening mammalian gene expression plasmids within mammalian cell line development
  • Experience in designing CRISPR knock-out and ORF libraries for high-throughput functional genomics screens
  • Experience working safely with 3rd generation lentiviral plasmid systems in a CAT2.0 environment
  • Experience designing mammalian gene expression constructs for Immunotherapeutics molecules, such as antibodies
  • Experience working with bioprocess-relevant cell lines, like CHO or HEK293
  • Familiarity with mammalian cell line development, including GM/GE cell lines, single-cell cloning, and clonal screening
  • Experience with molecular biology techniques, including PCR, qRT-PCR, Western Blot, ELISA, plasmid preparation, plasmid cloning, site-directed mutagenesis, and FACS
  • Experience working with SnapGene or similar software
  • Skilled in experimental design and troubleshooting
  • Able to work independently and as part of a team
  • Good numeracy, computer, and communication skills
  • Experience writing SOPs and risk assessments

Salary: £40k - £55k + Benefits
