Previous experience in ES cell culture and gene targeting in ES cells is an advantage but not essential. Expertise in standard molecular biology techniques (PCR, sequencing, or _in-vitro_ transcription) are also desirable. Training in all skills including dissection will be provided.

**Main Duties/Key Responsibilities**:
ES cell culture and manipulation. Extraction of mouse tissues. Characterisation and quality control of genetically modified materials employing PCR- and ddPCR- based techniques and Nanopore sequencing. Genotyping. Gene expression studies. Sample management and communication of results.

Other general duties required are to ensure the smooth running of a molecular biology laboratory i.e., ordering reagents - tasks shared with the rest of the team.

***Working Relationships**:

- Process samples/tasks by working as a team with the other Molecular and Cellular Biology (MCB) team members, as directed by the MCB Deputy Head initially or by the Head of the Genotyping Core, occasionally
- Work with scientists in the different research groups, to help process samples as efficiently as possible

**Additional Information**:
Some flexible working patterns and paid weekend work may be required at the discretion of the supervisor.

Person Specification

**Education/Qualifications/Training Required**:

- Essential:_
- An understanding of basic techniques (e.g., qPCR, cloning, aseptic technique)
- Desirable: _
- An understanding of genetic modification in laboratory animals
- MSc in Biological Sciences with several years’ laboratory-based molecular biology training
- An understanding of genome-engineering techniques (e.g., gene targeting, CRISPR-based tools)

**Previous Work Experience Required**:

- Essential: _
- Previous work in aseptic conditions and/or using PCR-based techniques
- Desirable:_
- Previous experience of working with genetically modified materials
- Previous experience of working with ES cells or of digital PCR
- Previous experience in genotyping would be desirable, although some training in molecular techniques and automated liquid handling can be given. Evidence of quality and customer-focused work.

**Knowledge and Experience**:

- Essential: _
- Experience in one of the following molecular and cellular biology techniques: ES cell culture and gene targeting, PCR, Sanger sequencing analysis, ddPCR, in vitro transcription
- Computer literacy is essential for the recording and communication of results
- Desirable: _
- Working and adhering to standard operating procedures and experience of process analysis and refinement
- Experience in service provision

**Personal skills/Behaviours/Qualities**:

- Organised and hard-working, with a keen eye for detail and able to work well as part of a team

- Able to communicate clearly and concisely with a variety of scientists at all levels (from principal investigator to research assistants and animal technicians)
