Here at the world-famous and internationally respected Wellcome Sanger Institute, we have an exciting opportunity for an Advanced Research Assistant to join an interdisciplinary team on an ambitious project aimed to identify new genes involved in neurodegeneration, especially Alzheimer’s and Parkinson’s disease.

This is a 12-month fixed term contract covering maternity leave for an existing team member.

About the Role

As Advanced Research Assistant, you will:

- Use human induced pluripotent stem cell (hiPSC) derived microglia coupled with cutting edge high-throughput single cell ‘omics techniques to assay the effects of natural and induced genetic variation on gene expression and cellular phenotypes.
- Work under the leadership of Dr Andrew Bassett and Dr Gosia Trynka and with collaborators at the Target Discovery Institute and Dunn School of Pathology at the University of Oxford (Daniel Ebner and Sally Cowley) and industry partners including Glaxo Smith Kline, Bristol Myers Squibb, Sanofi and Pfizer.
- Be able to take advantage of the exceptional resources and experience at the Wellcome Sanger Institute and more broadly within Open Targets in human disease genetics, iPSC-based cellular modelling, high throughput CRISPR screening and single cell ‘omics analysis.

About You

Cell culture and molecular biology experience, and an ability to work both independently and as part of a team are essential.

Essential Skills
- MSc or BSc in biological sciences with 2-3 years relevant laboratory experience
- Extensive experience culturing primary and/or cell lines
- Ability to design and troubleshoot experiments
- Interest in neurodegeneration, stem cell biology, _in vitro_ differentiation and/or cellular modelling of disease
- Theoretical knowledge of CRISPR based genome engineering techniques e.g. knockout, knock-in, arrayed and pooled screening
- Theoretical knowledge of molecular biology strategies and techniques e.g. nucleic acid purification, PCR, qRT-PCR, Illumina library preparation
- Excellent organisational skills, capable of working to a schedule and deliver projects within agreed deadlines.
- Self-motivated, with an ability to work both as part of a team and also independently once appropriate training and guidance have been provided
- Good oral and written communication skills to allow accurate information to be exchanged.

Ideal Skills
- Experience of human ES or iPS cell culture
- Experience of _in vitro_ cellular differentiation especially to macrophages or microglia
- Experience of phenotypic assays (e.g. phagocytosis, cytokine treatment or release, migration, etc.)
- Practical experience in CRISPR based genome engineering techniques e.g. knockout, knock-in, arrayed and pooled screening.
- Experience with generation and analysis of genomics datasets (e.g. RNA-seq, ATAC-seq, ChIP-seq) especially single cell ‘omics.
- Good computer skills in the use of office software (e.g. Excel, Word, Powerpoint).

Other information

The cellular and gene editing research group (Bassett) is interested in the development of genome editing techniques, cellular differentiation and cellular phenotyping systems, especially with respect to high-throughput investigation of gene and non-coding regulatory element function in neurodegenerative disease. The immunogenomics group (Trynka) aim to understand the mechanisms through which genomic variants act to disrupt critical molecular pathways in the human immune system that cause disease with a goal to map genetic variants to functional cellular effects.
