Role Summary
 
The Synthetic Genomics group within R&D at Autolus is responsible for generating the synthetic plasmid DNA constructs encoding the chimeric antigen receptors that are expressed on the surface of CAR T-cells. We are looking to recruit a Research Technician who will be involved in plasmid DNA assembly, purification and sequencing. This is a lab-based role requiring an individual with molecular biology experience who is familiar with cloning techniques, including Golden Gate, Gibson and fusion PCR, and Sanger sequencing and next generation sequencing (NGS). Highly motivated individuals who would like to contribute to the development of cutting-edge CAR T cell technologies are invited to apply for this role.

Key Responsibilities
 
•    Purifying plasmid DNA (Miniprep to Gigaprep)
•    Amplifying DNA by PCR
•    Digesting DNA fragments using restriction endonucleases
•    Purifying DNA fragments
•    Assembling plasmids using Golden Gate, Gibson or fusion PCR
•    Sequencing plasmid DNA and interpreting results
•    Archiving plasmid DNA
•    Writing up cloning results in electronic lab book
•    Designing plasmids and primers
•    Assisting in the maintenance of laboratory equipment and reagent supplies
•    Ensuring compliance with laboratory protocols and safety guidelines.
•    Participating in lab meetings.

Experience

•    BSc or equivalent in Molecular Biology/Biochemistry or related discipline (E)
•    Knowledge and experience molecular cloning (E)
•    Strong verbal and written communication skills (E)
•    Good attention to detail (E)
•    Ability to identify problems and implement solutions (E)
•    Commitment to high quality work (E)
•    Ability for creative and critical thinking (D)
•    Experience of working in a busy laboratory environment is desirable, although full training will be given (D)
