Position Marie‑Curie PhD Studentship – ON‑Track (Fixed Term) at University of Cambridge Location: Cambridge, England, United Kingdom Project Overview This PhD studentship focuses on protein engineering of enzymes for in vitro directed evolution and machine‑learning based elaboration of biocatalysis for oligonucleotide synthesis. The research is part of the Horizon Europe EU Marie Curie Network ON‑TRACT (www.on‑tract.eu) and involves collaborations with EU and UK groups engaged in therapeutic oligonucleotide development. Responsibilities Develop ultrahigh‑throughput platforms for in vitro enzyme evolution relevant to synthetic chemistry, including chiral synthesis (IREDs, ligases). Create diverse libraries via error‑prone PCR, site‑saturation mutagenesis, and InDel transposon‑based methods. Conduct in vitro selections, long‑read nanopore sequencing and deep mutational scanning of screening outputs. Analyze sequence‑function datasets using regression and tree‑based machine‑learning algorithms to generate improved mutants and mechanistic insights. Collaborate across the ON‑TRACT network, gaining training in organic synthesis, cell biology, medical work, and other complementary skills. Qualifications First or upper‑second class degree in chemistry, biochemistry or a related field followed by a PhD. Familiarity with molecular and chemical biology techniques and specific skills in assay development, enzyme kinetics, DNA library generation, structural biology, organic synthesis, high‑throughput screening, mechanistic enzymology, bioinformatics, and microfluidic engineering are advantageous.
