**Clinical AMN2 Scale - £43,504 - £64,461**

**Deanery of Clinical Sciences/Institute for Regeneration and Repair/Centre for Inflammation Research**

**Contract Type - Fixed Term - 3 Years**

**Full Time - 35 Hours Per Week
A Clinical Fellow position is available for a three-year Research Fellowship (to count towards a PhD higher research degree) with Professor Neil Henderson within the Centre for Inflammation Research at the University of Edinburgh.

**The Opportunity**:
Currently, therapeutic options for patients with chronic liver disease are limited to removal of the underlying cause, if possible, and transplantation reserved for a select few. Importantly, the degree of liver fibrosis predicts adverse clinical events including clinically significant portal hypertension, hepatic decompensation and the development of hepatocellular carcinoma. Therefore, developing effective anti-fibrotic therapies for patients with chronic liver disease is likely to impact significantly on morbidity and mortality, and hence, the discovery of effective anti-fibrotic therapies remains one of the holy grails in hepatology.

Single-cell RNA sequencing (scRNA-seq) has the potential to deliver a step change in both our understanding of healthy liver tissue homeostasis as well as disease pathogenesis, such as NASH-induced liver fibrosis and cirrhosis, allowing the interrogation of individual cell populations at unprecedented resolution. In this project, the Fellow will use and be trained in a broad range of cutting-edge single cell genomics techniques including single cell and single nuclei RNA sequencing (10X Genomics Platform) of freshly isolated cells from human liver tissue (uninjured and explant tissue obtained at time of liver transplantation), spatial transcriptomics, multi-colour flow cytometry/cell sorting, confocal microscopy, and human liver fibrosis imaging approaches (eg PET-based Tracer / MRI liver imaging in patients with chronic liver disease).

Application of new single cell genomics and PET-based imaging approaches in chronic liver disease should greatly facilitate the discovery of novel, rational therapeutic targets and pathways to treat patients with liver fibrosis. This work should also inform molecular pathology-based patient stratification, which is fundamental to the prosecution of successful clinical trials in human liver fibrosis.
